does target means the local google domain or the search location?

in Find new keywords and get search volume data,

When targeting specific country, does it imply users who are searching using the local google of the country, or everyone searching from that country?

Example, if I select Canada as the target, does it mean it will count only searches done through google.ca or will count all searches done from Canada regardless if google.com or google.ca is used?

It will count all searches performed in Canada.  I believe it will also count searches performed on google.ca from people physically outside of Canada because it implies specific interest in Canadian results.

If you click on Location options (advanced) in setup you have three options:

People in, or who show interest in my targeted location (recommended)
People in my targeted location
People who show interest in my targeted location

So in your case if you select "People in my targeted location" it will be anyone in Canada. You must keep in mind Google uses search partners as well. Is there a reason you'd only want to show ads to people doing searches done through google.ca?
